Scientific Overview

Alpha-Klotho is a single-pass transmembrane protein with a shed soluble form that acts as a circulating hormone. Klotho-overexpressing mice live ~30% longer; klotho-deficient mice age prematurely. Dena Dubal's lab (UCSF, 2014, 2020) showed that exogenous soluble klotho improves cognition in aged mice and primates. Human trials remain limited.
