Growth Hormone-Releasing Hormone
GHRH is the hypothalamic peptide that signals the anterior pituitary to release growth hormone in pulsatile bursts. The native molecule is 44 amino acids long, but the bioactive fragment is the first 29 residues — which is why most synthetic GHRH analogs (sermorelin, tesamorelin, CJC-1295) are 29-residue peptides with structural modifications added for serum stability.
In a research context, GHRH analogs are investigated because they preserve the body’s feedback loop on GH release. Unlike exogenous somatropin, which floods the system with HGH directly, a GHRH analog asks the pituitary to release its own GH — which means the hypothalamic-pituitary axis can still down-regulate the response if endogenous IGF-1 climbs too high. That self-limiting property is the central reason GHRH-class peptides are popular in long-running protocols.
GHRH analogs are commonly stacked with a GHRP (Growth Hormone-Releasing Peptide) such as ipamorelin or hexarelin. The two classes work on different receptors — GHRH on the GHRH-R and GHRPs on GHS-R — and the synergy between them produces a larger GH pulse than either compound alone.
